A leptin-mediated central mechanism in analgesia-enhanced opioid reward in rats.

چکیده
Opioid analgesics are commonly used in chronic pain management despite a potential risk of rewarding. However, it remains unclear whether opioid analgesia would enhance the opioid rewarding effect thereby contributing to opioid rewarding. Utilizing a rat paradigm of conditioned place preference (CPP) combined with ankle monoarthritis as a condition of persistent nociception, we showed that analgesia induced by either morphine or the nonsteroid anti-inflammatory drug ibuprofen increased CPP scores in arthritic rats, suggesting that analgesia itself had a rewarding effect. However, arthritic rats exhibited a significantly higher CPP score in response to morphine than ibuprofen. Thus, the rewarding effect of morphine was enhanced in the presence of persistent nociception, producing a phenomenon of analgesia-enhanced opioid reward. At the cellular level, administration of morphine activated a cascade of leptin expression, glial activation, and dopamine receptor upregulation in the nucleus accumbens (NAc), while administration of ibuprofen decreased glial activation with no effect on leptin expression in the NAc. Furthermore, the morphine rewarding effect was blocked in leptin deficient ob/ob mice or by neutralizing leptin or interleukin-1β in the NAc without diminishing morphine analgesia. The data indicate that systemic opioid can activate a leptin-mediated central mechanism in the NAc that led to the enhanced opioid rewarding effect. These findings provide evidence for an interaction between opioid analgesia and opioid rewarding, which may have implications in clinical opioid dose escalation in chronic pain management.

Chronic pain is a prevalent and debilitating condition, conveying immense human burden. متن کاملPost-stress analgesia after lesions to the central nucleus of the amygdala in rats.
In two experiments the role of the central nucleus of the amygdala under two modes of foot-shock analgesia was studied in 39 male Möll-Wistar rats. In Experiment I a 4 min continuous foot-shock, dependent on neural mechanism was used as a stressor. Analgesia was produced by regularly intermitted 20 min of foot-shock action in Experiment II which evoke an opioid, humorally mediated mechanism. Th...
متن کاملStudy on the possible similar mechanism of ultra low dose-induced hyperalgesia and development of tolerance to analgesia in male rats: an study based on the role of Gs signaling pathway
Introduction: Ultra low dose (ULD) morphine induces hyperalgesia which is mediated by excitatory Gscoupled opioid receptors. This study was designed to investigate the development of tolerance to hyperalgesic effect of morphine. Also we attempt to seek possible similarity, in view of Gs proteins, between hyperalgesic effect of ULD and hyperalgesic effect after tolerance to HD. Method: Male ...
متن کاملCentral serotonergic neurons are differentially required for opioid analgesia but not for morphine tolerance or morphine reward.
Opioids remain the most effective analgesics despite their potential adverse effects such as tolerance and addiction. Mechanisms underlying these opiate-mediated processes remain the subject of much debate. Here we describe opioid-induced behaviors of Lmx1b conditional knockout mice (Lmx1bf/f/p), which lack central serotonergic neurons, and we report that opioid analgesia is differentially depe...
متن کاملOpioid-induced glial activation: mechanisms of activation and implications for opioid analgesia, dependence, and reward.
This review will introduce the concept of toll-like receptor (TLR)-mediated glial activation as central to all of the following: neuropathic pain, compromised acute opioid analgesia, and unwanted opioid side effects (tolerance, dependence, and reward). Attenuation of glial activation has previously been demonstrated both to alleviate exaggerated pain states induced by experimental pain models a...
